| Summary: | 碩士 === 國立高雄海洋科技大學 === 水產食品科學研究所 === 101 === 2, 3, 5, 4'- tetrahydroxystilbene-2-O-beta- D-glucoside (THSG) is an major component of Polygonum multiflorum Thunb (Ho-Shou-Wu). This study aims investigate regulation of THSG in colon cancer cell (HT-29 cells) metastasis, and effects of THSG on colon cancer forming and metastasis on F344 rats induced by azoxymethane (AOM), respectively. According to results of cell model experimentals, when HT-29 cells treated 10 mM THSG were significantly increased wound-healing area percentage of HT-29 cells after 24, 48 or 72 hrs, respectively, incubation as compre control group (p<0.05), . 1, 5, 10 mM THSG groups were significantly lower on protein level of metalloproteinase-2 (MMP-2) than control group after 24 hrs incubation (p<0.05). These results show that THSG could suppress the migration of HT-29 cells. From adhesion reaction assay results show that 1, 5, 10 mM THSG were significantly decreased HT-29 cells adhesion to EA. hy926 cells as compare control group (p<0.05). Endothelial cell selectin (E-selectin) and intercellular adhesion molecule-1 (ICAM-1) protein levels of EA. hy926 cells were significantly decreased after THSG treatment (p<0.05). These results show THSG could inhibit the cell adhesion in metastasis of colon cancer. THSG also is a potential inhibit agent on cancer cell invasion. Because, invasion assay results indicated 5 and 10 mM THSG could inhibit HT-29 cells invasion as compare control group (p<0.05). 5 and 10 mM THSG also could significantly increase the trans epithelial electric resistance (TEER) of EA. hy926 cells (p<0.05). And, 10 mM THSG was significantly decrease phosphorylates vascular endothelial cadherin (p-VE-cadherin) protein expression (p<0.05). These results demonstrate the anti-metastatic properties of THSG. In addition, from animal experiments results show, 30, 150 or 250 mg/kg BW THSG treatment for 6 wks do not effect on growth characteristics on AOM-induced F344 rats. From aberrant crypt foci (ACF) number analysis, ACF number were significantly increased in AOM control group as compare control group rats (p<0.05). But, ACF number is significantly descreased on 30, 150, or 250 mg/kg BW THSG group rats after 6 wks treatment. Immunohistochemical staining study results show the protein expression of CEA were significantly descreased after 30, 150 or 250 mg/kg BW THSG treatment for 6 wks as compare AOM control group rats (p<0.05). In conclusion, THSG were a potential compound on human colon cell line (HT-29 cells) through suppression migration, invasion and invasion of metastasis. And, THSG can inhibit F344 rats colon cancer forming induced by AOM. But, effects of THSH on colon cancer metastasis on rat needs a further research.
