It has been reported that enterovirus protein, 3A, is know to be required for formation of replication vesicles, and mutations in 3A give rise to defects in viral RNA synthesis. In the present study, we found that full length enteroviral RNA with deletion of 3AB can not replicate and results in dead-end infection in RD cells. We have constructed recombinant plasmids for expressing the 3AB protein and its variant fusion proteins. The expressed 3AB proteins are readily processed by viral proteases accompanied with the following EV71 infection to the transfected cells. Our current result implicates that the expressed 3AB protein may rescue the lethal 3AB deletion mutant by trans-complementation. Defective infectious viral particles could therefore be produced in a 3AB stably expressing cell line. Since the defective particles can not replicate in normal cells, 3AB deletion is presumed to be a universal attenuation strategy for generating vaccine strains for enteroviruses.
