Summary: | 博士 === 國立嘉義大學 === 應用化學系研究所 === 101 === In this study, mono-substituted resveratrol derivatives containing multi-carbon chain (RMS) are used to form liposomes by ethanol injection /sonication method, RMS liposomes are observed in different storage temperature(4℃, 25℃) and time (0 ~ 168 hr) .The sizes and shapes of RMS liposomes are monitored by following instrument : particle size distribution analyzer, transmission electron microscopy (TEM) and energy dispersive spectroscopy(EDS) .The average particle size of RMS liposomes stored at 4℃ and 25℃ after 0 ~ 168 hr are 223 nm~ 238 nm (4℃)and 255 nm ~ 265 nm(25℃) . The result shows that temperature is a factor to affect particle size and lipid interactions, which result in instability and therefore make the particle size grow larger. In TEM spectroscopy, the RMS liposomes become double helical structure after 8 hr. To understand the physical property of the double helical structure, the RMS liposome is tested by following condition: illumination process, Urea addition and different pH buffer addition. The illumination test shows the RMS liposome become double helical structure of non- chirality in the R form and S form after 2 hr of fluorescent lamp illumination, and the illumination process will accelerate the formation of helical structure. On the other hand, by exposure to the UV light, the microsphere structure collapse because the energy of UV light is greater than fluorescent lamp. The Urea adding test shows that there is no helical molecule after adding in room temperature for 1 hr. Its presumably due to the hydrogen bonding of the helical structure. After 24 hr, the helical structure is still unformed. The pH buffer adding test shows that there is no helical structure after adding pH 6, pH 7 and pH 8 buffer to the RMS liposome for 48 hr. For the application of RMS liposome, the drug controlled release ability is tested through nano gold, ruthenium complexes and DNA and other drugs. The results shows RMS / Au liposomal formulations at 4 ℃, 25 ℃ and 37 ℃ for 48 hr to form a double helix, and the release of gold nanoparticles and ruthenium complexes. According to the experimental results, the drug release ability of RMS liposome double helix molecule can be controlled, but the structure is self-assembled into tiny balls after 168 hr. Resveratrol has antioxidant activity; therefore, the derivatives and RMS liposomes are tested for its antioxidant activity separately. The experimental result shows resveratrol derivatives (RMS) and liposome composed of RMS can scavenge free radical for 59% to 61%, indicating that resveratrol derivatives (RMS) antioxidant activity will not decrease in the formation of derivatives and different structures. Because of the antioxidant activity of RMS, the RMS / dye (hematoxylin) liposomal formulations and hair care formulations used in hair dye, and conduct anti-UV test. By the RMS/dye liposomes hair dye formulations tested, dyed hairs are washed with water and apply conditioner RMS liposomal formulations and EPC liposomal formulations, after 60 hr of exposure to ultraviolet light to detect spectral color measurement instrument. The results shows the uncoated hair bundle is 4.71 ~ 7.32 △ E; the hair bundle after treating RMS △ E value is 4.05 ~ 4.64. It shows the RMS liposome formulations can protect hair from UV irradiation.
