Description
Summary:碩士 === 國立中興大學 === 生命科學系所 === 101 === Flavonoids are mainly in the grains and herbs. In the West, it is estimated about 20-50 mg flavonoids intake a day. In recent years, people are more interest in flavonoids because they believe that flavonoids can inhibit atherosclerosis, osteoporosis, diabetes, and some cancers. Flavonoids have been found in many plants, food and tea. Many people believe that flavonoids in the human body have diversity role. In general, flavonoids may act as an antioxidant let free radical expelled from the body. In addition, flavonoids may affect estrogen increase, this may be due to inhibition of the aromatase enzyme production - an important estrogen biosynthesis enzyme. For these reasons, flavonoids are thought to reduce the incidence of breast cancer, and prostate cancer. The three chemicals in this study : Chrysin, Baicalein and myricetin are flavonoids. We discuss the cytotoxicity in immune cell HL60 and Jurkat to expect to develop new drugs for the treatment of leukemia or other cancers in the future. The results illustrate that these drugs can cause cell death might be through to the high level ROS and decrease mitochondrial membrane potential, finally the endogenous mechanism of apoptosis initiated. Ornithine decarboxylase (ODC) catalyzes the first and rate-limiting step in polyamines biosynthesis. It plays an important role in cell cycle, cell growth, differentiation and apoptosis, also plays a role of tumor-promoting factor. Therefore, this study also explored the height of ornithine decarboxylase whether the performance can delay cell death. From the experimental results, overexpression of ornithine decarboxylase these three drugs can delay cell death caused.
