But as the study of disease mechanism is still unclear, this study is to investigate the relationship between fibrocytes and asthma, as well as the condition of its growth regulation. Methods: To set up the method of multiparametric flow cytometry to analyze percentages of fibrocytes in human peripheral blood, and examine the relationship between fibrocytes and clinical parameters in asthma patients. Using human recombinant TGF-β and TGF-β RI inhibitor SB 431542 to examine the signaling involved in fibrocyte differentiation and growth. Results: We defined CD3-CD19-CD45+Collagen+PM-2K- cells as fibrocytes, and found that the percentages of circulating fibrocytes in asthma patients were significantly higher than healthy controls, but macrophage percentages were significantly lower than healthy controls. In addition, fibrocyte percentages were negatively correlated with macrophage percentages and positively correlated with IgE level in asthma patients. In healthy controls, the differentiation and growth of fibrocytes from CD14+ monocytes were inhibited by TGF-β RI inhibitor SB 431542 and promoted by human recombinant TGF-β. Conclusions: This study contributes to an understanding for the association of fibrocyte in the disease activity of asthma.
