We recruited a total of 379 male subjects aged 20 to 25 participated in this thesis. Before collection the blood, all study subjects provided, informed consent and a self-report questionnaire about smoking behavior. Analysis the single nucleotide polymorphism (SNP) of dopamine metabolism genes (DRD2, COMT) with the methods of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Finally, the results of genetype and the questionnaire were performed statistical analysis. Odds ratio(OR) of smoking was 3.768 higher [95% confidence interval (CI):1.023~13.874, P=0.034] in the subjects carrying DRD2 TaqI A A1A1 (TT)/TaqI B non-B1B1 (GG, GA) than DRD2 TaqI A A1A1 (TT)/TaqI B B1B1 (AA) carriers. Physiological cigare dependence scale was significantly higher in the smokers possessing DRD2 TaqI A A1A1 (TT)/TaqI B non-B1B1 (GG, GA) than those possessing DRD2 TaqI A A1A1 (TT)/TaqI B non-B1B1 (GG, GA) [mean (± SD) 41.44 (± 17.38) versus 32.55 ( ± 13.42), P=0.042]. Fagerstrom test for nicotine dependence in the smokers carrying COMT rs4680 GA or AA/COMT rs165599 AA [4.47 ( ± 1.73)] was significantly higher than that [2.94 (± 1.72)] in those carrying COMT rs4680 GA or AA/COMT rs165599 AG or GG (P=0.005). When the current-smokers were divided into moderate/heavy smokers and light smokers, the OR of moderate/heavy smoking in the subjects carrying COMT rs4680 GA or AA/COMT rs165599 AA was 7.422 higher than those carrying COMT rs4680 GA or AA/COMT rs165599 AG or GG (95% CI:1.578~34.902, P=0.005). The results revealed that variation status of DRD2 TaqI A A1A1 (TT)/TaqI B non-B1B1 (GG, GA) and COMT rs4680 AA/COMT rs165599 AA allele are associated with smoking behavior in male Taiwanese young male adults.
