Because of the amino acid sequence mutants in active site of YKL-40, it has lost the ability to cleave chitin but still retain the ability to combine with chitin. Previous studies have found that expression of YKL-40 correlate with asthma severity and are considered a biomarker for asthma. CHI3L1 gene promoter -131C/G (rs4950928) single nucleotide polymorphism (SNP) influences transcription of YKL-40. CHI3L1 -131C/G SNP is proved to be associated with asthma. Most relative researches mainly focus on Caucasian previously. The research report focusing on Asian race indicates that the YKL-40 serum levels of asthma patients are higher than controls in Chinese adults and it is similar to previous studies. However, CHI3L1 -131C/G SNP is not associated with asthma in Korean children. It is totally different from the research done by western countries. This study aims to understand the relationship between CHI3L1 -131C/G SNP and asthma. We analyzed the data of CHI3L1 -131C/G SNP and YKL-40 serum levels taken from adult asthma patients and healthy adults in Taiwan. Then we provided these data to discuss widely and know the relationship between CHI3L1 -131C/G SNP and asthma. In addition, allergic rhinitis patients were added into this research. Materials & Methods: We recruited 183 subjects, including 70 asthma patients, 42 allergic rhinitis patients and 71 healthy controls. Peripheral venous blood was collected to examine eosinophil cationic protein (ECP), total immunoglobulin E, WBC count, WBC differential count and Eosinophil count. CHI3L1 -131C/G genotype was performed by using TaqMan® SNP genotyping assay. YKL-40 serum level was performed by using MicroVue™ YKL-40 EIA kit. Multiple sequence alignment was performed in 760 bp DNA sequences of CHI3L1 promoter region. Statistical analyses were performed by using SPSS software. Results: The result demonstrated that there were significant differences between asthma group and control group in total immunoglobulin E (p = 8.38×10-9), WBC count (p = 1.25×10-5), lymphocyte percentage (p = 5.97×10-4), monocyte percentage (p = 0.012), eosinophil percentage (p = 4.43×10-5) and eosinophil count (p = 7.82×10-8) data. There were also significant differences between allergic rhinitis group and control group in total immunoglobulin E (p = 1.60×10-8), eosinophil percentage (p = 1.64×10-6) and eosinophil count (p = 1.27×10-5) data. There was no significant difference between asthma group and control group in CHI3L1 -131C/G SNP data (p = 0.413), but there were significant differences between asthma group and control group in YKL-40 serum levels data (p = 2.10×10-3). There was no significant difference between allergic rhinitis group and control group in CHI3L1 -131C/G SNP and YKL-40 serum levels data (p = 0.987 and 0.068). Conclusion: In conclusion, YKL-40 serum levels are associated with asthma. The relationship between CHI3L1 -131C/G SNP and asthma seems to be different in Eastern and Western.
