Summary: | 碩士 === 中山醫學大學 === 生化暨生物科技研究所 === 101 === Obesity is a worldwide health problem, and it is a significant risk factor for various diseases such as diabetes, cardiovascular disease, hypertensive and cancer. However, obesity is mainly result of the increase volume due to fat, inhibit adipocyte differentiation and adipogenesis is the main way to evaluate anti-obesity. Related research report, rich in many natural plant phenolic compounds have been shown to have antioxidant, anti-inflammatory, anticancer and lower cholesterol properties. Therefore, this study hopes to find the compounds from food with anti-obesity ingredients, thus achieving the anti-obesity effect. One of the plants which are considered medicinal is Hibiscus sabdariffa Linnaeus with local name of Roselle. In this study, Roselle extract rich in polyphenols-anthocyanins, assess whether inhibition of adipocyte proliferation and the regulation of adipogenesis. Using the 3T3-L1 adipocytes cell model, we explored the effect of HAs (Hibiscus anthocyanin) on cell proliferation and adipogenesis in 3T3-L1 preadipocytes. Experimental studies include: MTT assay, WST assay, PI stain/FACScan, Oil red O stain, intracellular triglyceride accumulation assay and western blotting. These results suggest that HAs can effectively inhibit adipogenesis in 3T3-L1 preadiopocytes and adipocytes, also regulating the performance of PPAR γ and FAS。Therefore, Hibiscus anthocyanins may have potential for the treatment of obesity.
