Summary: | 碩士 === 嘉南藥理科技大學 === 環境工程與科學系 === 102 === The research is to establish Enzyme-linked immunosorbent assay (ELISA) for contaminant soil and apply on contaminant site of Dioxins. Phase one is to establish the standard operation procedure of ELISA, including drying soil, extraction, clean up and data analysis. Phase two is to apply ELISA on the contaminant site, meanwhile; to match the data with High resolution gas chromatograph/high resolution mass spectrometer (HRGC/HRMS).
The result shows that the concentration of Dioxins reaches the maximum in two hour of extraction. The concentration of Dioxins reaches the maximum from carbon column washed by 20 ml of Toluene. Moreover, in the part of application for the site, 12 of 78 samples have been analyzed by HRGC/HRMS for Calibration Adjustment Factor (CAF).After correction of CAF. The results shows that 9 samples analyzed by ELISA screening test appear positive, 7 of 9 samples analyzed by HRGC/HRMS also appear positive.
In the part of cost saving, if all samples analyzed by HRGC/HRMS replaced to ELISA screening test, the cost could saved by 975,000 TWD and 16 days of construction period.
ELISA screen test can analyze Dioxin sample quickly. A large number of samples screening by ELISA for the investigation in a large contaminant site can understand the distribution and trend of pollutant. If it’s necessary, the representative samples could be analyzed by HRGC/HRMS. ELISA is not only saving money and time but also is a benefit for investigation.
