Environmental carcinogens such as betel quid chewing, tobacco smoking and alcohol drinking have been identified as major risk factors for OSCC. In the past, our lab analyzed the genomic aberration of OSCC patients by array-CGH, we have found the amplification of the q21 band of chromosome 8 (8q21) occurs in a large percentage of OSCC. WW domain containing E3 ubiquitin protein ligase 1 (WWP1) was a potential oncogene, WWP1 was a ubiquitin ligase, associated with the post-translational regulation of several tumour-promoting and tumour suppressor proteins. WWP1 has previously been shown to be frequently amplified and overexpressed in OSCC. In this study, we investigate the function of WWP1 by RNAi in order to understand the molecular mechanismin OSCC tumorigenesis.Inhibition of cell growth was examined by Microscope and MTT assay. The effects on apoptosis were determined by flow cytometry. Our findings showed that knock-down of WWP1 would cause apoptosis in OSCC cell lines. We established the LacO/IPTG controlled shRNA inducible cell system to investigate the functional roles of WWP1 in OSCC. The apoptosis proteins such as caspase3、caspase7 and PARP are induced, p53 is suppressed by western blot. Our results reveal that WWP1 may play an important role in tumorigenesis of oral cancer.
