From our previous study, we found the inflammatory cytokine: interleukin-1β (IL-1β) stimulates mesenchymal stem cell migration by up-regulating myosin light chain kinase (MLCK) expression. PKC/MEK/ERK/NF-κB signaling transduction pathway is involved in IL-1β-induced MLCK expression, but the mechanism has not been fully understood. In this study, we show that IL-1β induces PKCα/βI/δ isoforms, MEK/ERK, and NF-κB signaling activation. By pretreatment with PKC inhibitors: Calphostin C, Go 6976, PKCβ inhibitor, Rottlerin on IL-1β-induced MEK/ERK phosphorylation, we found that PKCα acts upstream of MEK/ERK activation in IL-1β stimulation. Moreover, pretreatment with PKC inhibitors, MEK inhibitor U0126, ERK inhibitor PD98059 on IL-1β-induced IκBα phosphorylation, we found that PKCδ acts upstream of NF-κB signaling activation in IL-1β stimulation. We also demonstrated that NF-κB subunit p65 translocates to nucleus in IL-1β stimulation. In conclusion, IL-1β-induced mesenchymal stem cells NF-κB signaling activation is dependent on PKCδ and then NF-κB subunit p65 translocates to nucleus to up-regulate it’s expression.
