Developing a Rat Model of Tourette Syndrome for Screening Potential Lactic Acid Bacteria and Traditional Chinese Medicines on DOI-induced Stereotype Behavior Improvement

碩士 === 國立陽明大學 === 生化暨分子生物研究所 === 100 === Tourette syndrome is a neuropsychiatric disorder. It is characterized by sudden, involuntary and repetitive tics. It is characterized by sudden, involuntary and repetitive tics. Lactic acid bacteria (LAB) is a specie of bacteria that benefit human enteric system. In this study, a Tourette rat model induced by DOI ((±)-1-(2,5-Dimethoxy-4-iodophenyl)-2-aminopropanehydrochloride) is generated. The potential lactic acid bacteria and traditional Chinese medicines which can improve the DOI-induced stereotype behaviors will be searched with this model. The results showed that LAB 7036 (109, 1010 CFU/ml live bacteria)and Gan mai da zao tang (400mg/kg body weight) had better effects on reducing the DOI-induced back muscle contraction. According to the quantification of bacterial communities in caecal contents, the data suggested that the administration of live LAB 7036 would alter the gut microbiota of Tourette rats. In addition, LAB 7036 suppressed the expression of DOI-induced dopamine transporter (DAT) and dopamine D2 receptors (D2R) in the cortex and the striatum dose-dependently. In conclusion, LAB 7036 could improve the DOI-induced stereotype behaviors by suppressing the expression of DAT and D2R in the cerebral cortex and the striatum through the influence on gut microbiota of Tourette rats.
