During the fermentation of X. dendrorhous, zinc ion and nitrogen source as well as pH value of the culture broth strongly affect the cell grow and the yield of neo-FOS. Maximal neo-FOS (145 g/L) was obtained when the fermentation was carried out at pH 7 for 48 h, using 20%(w/v) as substrate. Addition of malt extract (3 g/L) enhanced the cell growth. When 30%(w/v) was used, addition of zinc sulfate heptahydrate (0.05 g/L) resulted in a higher amount of glucose compared with that without zinc ion, leading to a lower purity of neo-FOS. Culture with yeast extract (10 g/L) had faster cell growth than that with peptone (5 g/L). However, faster cell growth resulted in a lower purity of neo-FOS. Compared with stirred tank culture, air-lift tank culture produced more neo-FOS (165 g/L) and less amount of glucose, resulting in the highest purity of neo-FOS.
