Extracts of black queen grape winery waste have been shown to exhibit antioxidant and anti-inflammatory activities. This study was carried out to explore whether the fresh black queen grape had the same effects. The lyophilized fresh black queen grape skin or flesh samples were used to generate aqueous extract of fresh black queen grape skin (AGS), aqueous extract of fresh black queen grape flesh (AGF), ethanolic extract of fresh black queen grape skin (EGS), ethanolic extract of fresh black queen grape flesh (EGF). Their contents of total polyphenols, flavonoids and anthocyanins were determined. Antioxidant capacity was assayed by trolox equivalent antioxidant capacity (TEAC), DPPH free radical scavenging activity, and reducing power. The anti-inflammatory effects and the possible mechanisms were evaluated using the RAW264.7 macrophage cell model. Results indicated that the contents of polyphenols, flavonoids and anthocyanins were highest in EGS (12.34 mg/g, 3.95 mg/g and 1.65 g/mg, respectively). EGS also had the best antioxidant capacities as determined by TEAC, DPPH scavenging or reducing power. Both EGS and EGF effectively inhibited lipopolysaccharide (LPS)-induced productions of nitric oxide (NO), tumor necrosis factor (TNF)-α, interleukin (IL)-6 and IL-10, as well as the expressions of inducible NO synthase (iNOS) and cyclooxygenase (COX)-2 proteins in a dose-dependent manner. On the opposite, LPS-stimulated protein expressions of nuclear factor (NF)-κB and inhibitor of NF-κB (IκB)-α were dose-dependently increased by EGS and EGF. Taken together, these observations show that ethanolic extracts of fresh black queen grape skin and flesh have antioxidant and anti-inflammatory capacities. The anti-inflammatory mechanism may be partially associated with suppression of NF-κB activation, inflammatory mediators NO, TNF-α, and IL-6 productions, as well as iNOS and COX-2 protein expressions.
