Description
Summary:碩士 === 慈濟大學 === 醫學檢驗生物技術學系醫學生物技術碩士班 === 100 === Antibiotics have been widely used in the prevention and treatment of bacterial infections. However, with the overuse of antibiotic, it results in the emerging of the multidrug-resistant bacteria, such as the multidrug-resistant Acinetobacter baumannii (MDRAB). Therefore, searching for new strategies to combat multidrug-resistant bacteria becomes a major challenge. To our knowledge, phage therapy can be one of the possible solutions. The isolated phage?淲B3 could infect the MDRAB strain M68630 and generate a turbid plaque. Under the electron microscope, two types of phage morphologies similar to podoviridae and siphoviridae were observed. Our sequence result for phage?淲B3 showed two DNA contigs with the length of 64,863bp and 25,689bp, respectively. Some experiments were performed to help clarify the questions raised from the sequencing results. In addition, various bioinformatic tools were used to analyze and annotate the contigs obtained. Also, the ORF encoding the putative tail tape measure proteins was successfully cloned, and expressed. In the hope that the results revealed from this work can benefit the control of the infections caused by MDRAB in the future.
