Description
Summary:碩士 === 慈濟大學 === 醫學資訊學系碩士班 === 100 === Mutations can affect the activation of tumor suppressor gene, due to the damage of DNA-repairing mechanisms. Thus, the continued DNA-damage accumulates and leads to cancer. The main functions of tumor suppressor gene protein are inhabitation of cell growth, regulation of mitosis and cell replication process. Mutation may bring significant changes of protein stability, which lead to loss the normal functions of proteins. Because tumor cells are sensitive to heat, they can be killed by heating easily without harming normal tissue. Through the study of protein stability may understand the mechanism between heating and tumor cells. The point mutations data were downloaded from the COSMIC ( The Catalogue of Somatic Mutations in Cancer ) and SNP ( single-nucleotide polymorphism ) from NCBI. We selected the largest mutation frequency of highly point mutations genes from COSMIC. To calculate the scores of protein by AAindex3 contact potential matrix. This study assumes that some specific genes such as protein score of oncogene and tumor suppressor gene mutation would give different distributions. We can identify the cancered person and healthy person by proteomics analysis. In the future, we hope the gene sequences can be apply to the protein stability calculations so that the amount of changes and the scores can distinguish the cancered people and estimate cancer risk.
