Summary:碩士 === 慈濟大學 === 醫學檢驗生物技術學系醫學生物技術碩士班 === 100 === This study developed two simple and environmentally friendly sample preteatment methods combined with gas chromatography-mass spectrometry for the determination of amphetamines and ketamines in urine samples. One is liquid-liquid extraction using ethyl acetate as extraction solvent and acetic anhydride as derivatization agent, the other is emulsification microextraction with organic droplet followed by acetic anhydride derivatization. The extraction solvent used in the latter method is of low density, low volatility, low toxicity and proper melting point around room temperature. The extractant droplets can be collected easily via centrifugation, and solidification at a lower temperature. Parameters that affect the extraction and derivatization efficiency, including buffer concentration, pH, centrifuging speed, volume of derivatization reagents, derivatization temperature and time, were optimized. Under the optimum conditions, the limit of detections (LODs), linear ranges, precisions (relative standard deviations; RSDs) and accuracy (relative errors; REs) of the two methods were evaluated for each analytes in this study.
