Our laboratory studies adipogenesis in cell culture models.
Screening of mouse kinase shRNA library showed that MAP4K3 (mitogen-activated protein kinase kinase kinase kinase 3) knockdown enhanced adipogenesis. 3T3-L1 cells can be morphologically transformed into adipocytes without 3-isobutyl-1-methylxanthine (MIX) in the differentiation medium after MAP4K3 knockdown. Both basal and insulin stimulated glucose uptake rate increased after MAP4K3 knockdown in 3T3-L1 adipocytes and qPCR showed expression of CBP300 and PCAF decreased. These results showed the important roles of MAP4K3 in adipogenesis and insulin response. Further studies will help elucidate the mechanism of MAP4K3 inhibition of insulin sensitivity. studies
