The leaf, flower, and stem of Ajuga bracteosa were extracted by hot water, ethanol, and methanol. Quantitative and qualitative analysis of polyphenolics contained in plants extracts were carried out by RP-C18 RP–HPLC using UV detector revealed six active phenolic compounds (gallic acid, protocatechic acid, caffeic acid, p-coumaric acid, rutin, and quercetin). The analysis of phenolic compounds are Total phenolic content (TPC), total flavonoids content (TFC) and tannin contents. Moreover, the antioxidant activity was evaluated with 2.2-diphenyl-1-picrylhydrazyl (DPPH), Trolox equivalent antioxidant capacity (TEAC), and reducing power assay. The antiproliferative effect was determined in a human hepatoma cell line (HepG2). These results revealed that A. bracteosa flower in hot water extracts contained high amounts of phenolic, flavonoids, and tannin at 35.27 mg/g (GAE), 14.28 mg/g (QE), and 8.04 mg/g (TE), respectively. Flower extracts by hot water of A. bracteosa showed the highest antioxidant activity analyzed by TEAC and reducing power assays. For the DPPH assay, leaf and flower part of A. bracteosa in hot water extracts exhibited the highest antioxidant activity. A. brcteosa flower extracts revealed the highest in antiproliferative activity. From correlation analysis, radical scavenging activity of A. bracteosa may be influenced by all phenolic content, while antiproliferative effect may due to flavonoid content. In conclusion, hot water extracts from flower of A. bracteosa might be used as a potential source of natural antioxidants and an anti-tumor agent.
