It has been used widely used in aquaculture industry as fungicide and ectoparasiticide in cultured fish eggs, fingerlings and adult fish since 1930s. However, malachite green and its metabolite leucomalachite green have been found to have several toxicological effects on both microbial and mammalian cells, including carcinogenic, mutagenic, chromosome breakage, tetratogenic effects and respiratory toxicity. Therefore, malachite green is not authorised for use in food producing animals in several countries. But, malachite green is still used in many parts of the word due to lack of a proper alternative. The aim of this study was to investigate the absorption of malachite green in tilapia after exposed to 2 ppm of malachite green solution, and the distribution of malachite green and its metabolite leucomalachite green in the tissues of tilapia. Moreover, the persistence of malachite green and leucomalachite green also were investigated in both short-term (0~48 hours) and long -term (0~84 days) rearing experiment. After 1 hour exposed in 2 ppm of the malachite green and leucomalachite green were found in every tissues of tilapia, and the average concentration arrived at 2.4 ppm of whole body. It was showed that malachite green was quickly absorption into fish tissues. And it also found that 66.1% of malachite green was rapidly transformed to leucomalachite green. After exposure, fish were transferred into glass aquaria with a continuous flow of dechlorinated tap water. The total content of malachite green and leucomalachite green was decreased quickly to 36% at 48 hours after exposure. And then, the total content of malachite green and leucomalachite green was decreased slowly to 3% at 84 days. However, it is also found at 84 days after exposure, the highest average residue level of leucomalachite green was found in bone, eyes and lipid, their average concentration were 2,486 ppb, 1,170 ppb and 3,490 ppb respectively.
