Small molecule compounds are commonly used in anti-tumor chemotherapy recently. The overall objective of this study is to elucidate the mechanism of Taiwanin A, a small molecule compound, isolated from Taiwania cryptomerioides Hayata, for the apoptosis of human lung carcinoma. Nonsteroidal Anti-Inflammatory Drug-Activated Gene-1 (NAG-1) is a tumor suppressor gene and belongs to the superfamily of TGF-β. Our preliminary data demonstrated that the treatment of Taiwanin A increased the expression of NAG-1 and subsequently led to the apoptosis of human lung carcinoma cell lines A549 and H460. Silencing the NAG-1 mRNA expression in A549 could reduce the cell toxicity caused by Taiwanin A addition. The experimental results showed that the JNK/SPAK pathway played a key role in the activation of NAG-1. The addition of the inhibitor of JNK pathway, SP600125, resulted in the reduction of NAG-1 expression as well as the loss of sensitivity of human lung carcinoma cells to Taiwanin A. At last, the results of xenograft tumor animal model treatment and immunhistochemestry showed that Taiwanin A could repress the tumor development in nude mice through similar mechanism. These data support the conclusion that Taiwanin A inhibits lung carcinoma growth by the activation of JNK pathway and the increased expression of NAG-1 both in vivo and in vitro.
