This study takes cell (human chondrocytes, SW1353) to be cultured in the biomaterial PDMS substrate; a microscope with CCD and PCI image card are used to show the images of cell morphology to be observed on the video camera. Specific single cell is selected first, then three kinds of different concentration (0.1%, 0.15%, 0.2%) of trypsin are added, and three kinds of shape in electrode design (convex, wavy, serrate) of dielectrophoresis (DEP) force is applied to induce the cells detach from the substrate. This study will try to find the optimal conditions and methods of trypsin concentration or DEP force to detach the cell. In this study, we find that the better condition of trypsin concentration is 0.2% and the detachment rate of cell is 29.03%. The fastest cell detachment time is the dielectrophoresis force due to the wavy shape electrode. But the optimal conditions and methods is the serrate shape electrode of dielectrophoresis force which the cell detachment rate is the highest about 83.33%. However, it also owns the highest survival rate without lysis about 60%.
