Allergic diseases not only interfere with individual health-related quality of life, but also make societal economic burden. To find resolutions of allergy therapy are under intensive investigations. Current clinical corticosteroid therapy in relieving allergic symptoms has many potential complications. Litchi (Litchi chinensis Sonn.) is a fruit that largely grown in Taiwan, and its pericarps and seeds are rich in phenolic content. Phenolic compounds have many biological activities such as anti-inflammation, anti-cancer, and anti-viral activity, and hepatic protection. The current study was designed to explore the ameliorative function in allergic symptoms. Rat basophilic leukemia cell line (RBL-2H3) was to in vitro screen the effects of litchi pericarp and seed extractions on mast cells degranulation induced by the ionophore, ionomycin stimulation. The release of β-hexosaminidase, histamine, productions of cellular reactive oxygen species (ROS), cellular calcium influx, and expression of IL-4 and TNF-α mRNA were assessed. Treatment with litchi pericarp and seed fractions markedly inhibited the release of β-hexosaminidase, histamine release, intracellular (ROS), cellular calcium influx ,and reduced the production of IL-4 and TNF-α mRNA. In addition, the effects of litchi extracts in ameliorating ovalbumin (OVA)-induced allergic asthma in a mouse model was conducted. In this study, paw edema, cell count and IgE level in the bronchoalveolar lavage fluid (BALF), IgE level serum from ovalbumin (OVA)-sensitized and -challenged mice were examined. The results showed that pretreatment of the animals by gavage litchi pericarp or seed extracts fractionations were effectively in inhibiting allergic oedema. Inflammatory cells such as neutrophils infiltrated into lung lavage were attenuated. In addition, a significant reduction in IgE level in serum was also confirmed. The current results suggest the ameliorative effects of litchi pericarp and seed extracts in allergic symptoms.
