Establish Polymorphic STR Marker's Gene Bank and Genic Polymorphism Analysis on Taiwan Dogs

碩士 === 國立嘉義大學 === 獸醫學系研究所 === 100 === Taiwan Dogs are recognized on a provisional basis by the FCI (Fédération Cynologique Internationale, FCI) in 2004. In 1980, a cooperative study on the subject of native Taiwan Dogs be asked endangered.Identify of breed and avoid hybridization is an important subject of restoration. To investigate the current status of Taiwan Dogs in Taiwan, we collected the blood from 57 Taiwan Dogs in Taoyuan (n=15), Taichung (n=16) and Pingtung (n=26) counties. To validate polymorphic microsatellite markers for parentage test, microsatellite loci analysis were conducted for this study. Overall genetic diversity in the three areas with close relationship : Number of effective alleles(Ne), Pingtung (5,692) >Taoyuan (4.1603)>Taichung (3.8617); Expected heterozygosity(HE ), Taoyuan (0.7846)> Pingtung (0.7781) > Taichung (0.7308). Accumulated Probability of Exclusion (PE) values, as an indication of marker validity for parentage tests: 0.999999562519. And finally, we found the facial phenotype with high close relation to genotype in Taiwan Dogs. In conclusion, this test system is work for restorstion, identity and parentage of Taiwan Dogs.
