This study is aimed at isolating fungal strain from fruiting body of Bovine Bolete (Suillus bovinus (Pers.) Roussel) produced at Slash pine stand, inoculating them to Slash pine and Taiwan red pine (Pinus taiwanensis Hay.) seedlings, and investigating the effects of Bovine Bolete on growth and physiological characteristics of Slash pine and Taiwan red pine seedlings.
The results revealed that the isolated strains cultured on PDA medium, MEA, medium and MMN medium, had the average growth rates of its strains were 0.32 ± 0.02 mm / day, 0.28 ± 0.02 mm / day and 0.18 ± 0.01mm / day, respectively. The results of morphological identification and DNA analysis confirmed that the isolated strain is Suillus bovinus belonging to genus Suillus, family Boletaceae.
The results revealed that roots of Slash pine appeared in dichotomous and simple forms, and of Taiwan red pine appeared in simple forms. Mantle and hartig net were found under optical microscope and scanning electron microscope. Inoculation test demonstrated that Bovine Bolete could symbiose with roots of Slash pine and Taiwan red pine seedlings and form ectomycorrhizae.
This fungal strain was inoculated onto roots of Slash pine and Taiwan red pine seedlings. 9 months after inoculation, the net height growth, net root collar diameter growth, total leaf area, leaf blades, specific leaf area, chlorophyll concentations, and photosynthetic rate of the inoculated seedlings of Slash pine were significantly higher than the non-inoculated ones. And the net height growth, total leaf area, leaf blades, specific leaf area, chlorophyll concentations, and photosynthetic rate of the inoculated seedlings of Taiwan red pine were significantly higher than the non-inoculated ones.
