An active process involving transdifferentiation of vascular smooth muscle cells into osteoblast-like cells, including changes in the expression of bone-associated and mineralization regulating proteins, rather than a passive process had been discovered in the past few years. Among the inducers of vascular calcification, phosphate is considered a major one. Magnesium plays an important role in mineral metabolism and vascular tone, however the role of magnesium in inhibiting vascular calcification had not been well studied. Therefore, we design the study to investigate the effects of magnesium on vascular calcification and also discuss the relationship between phosphate and magnesium transporters. We demonstrate that higher phosphate levels induce rat aortic calcification in a dose and time dependent manner, whether higher magnesium levels prevent vascular calcification, and the inhibitory effect is time dependent. We also find out magnesium may reduce vascular calcification through activation of transient receptor potential melastatin 7 (TRPM7) and suppression of type III sodium-dependent phosphate cotransporter Pit-1.
