To find a novel depigmentation agent, the anti-melanogenesis effects and possible whitening mechanism of Agaricus blazei murrill extract (ABE) were investigated. In this study, the antioxidant effects of ABE were in vitro determined by Folin–Ciocalteau assay, 2,2’-Azino-bis (3-ethylbenzo-thiazline-6-sulfonic acid) (ABTS) radical assay, 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, reducing power assay and metal chelating ability assay. Moreover, the inhibitory effect of the crude extract on mushroom tyrosinase activity, B16F10 melanoma intracellular tyrosinase activity and melanin content were also determined spectrophotometrically. The results indicate that ABE (0.1~0.75 mg/ml) decreased B16F10 intracellular melanin and decreased tyrosinase activity. Additionally, ABE also effectively decreased intracellular tyrosinase-related protein-1 (TRP-1) protein express in a dose-dependent pattern at the same dose range. Our results indicated that Agaricus blazei murrill extract show potent antioxidant activity and inhibit melanogenesis in B16F10 melanoma. Therefore, the crude extract may be effective when applied in depigmentation skin care products.
