With the change of life style, more refined eating and other pathogenic factors, cardiovascular diseases have become the top three of the ten leading causes of death in Taiwan, and killed millions of people every year. In the last few decades, Chinese herbs have been widely used in modern disease research. Scutellariae Radix has been found that it has anti-inflammatory, anticancer and antioxidant effects. Therefore, this research was to investigate the protective effects of Scutellariae Radix in cardiomyocytes.
In the research, the metabolite of Scutellariae Radix (50μL/mL and 100μL/ mL) was used to study the protective effects of Scutellariae Radix on hypoxia induced myocardial cells injury. The cells were placed to hypoxia chamber for 4 hours, 8 hours, 24 hours, 48 hours, 72 hours, and then reperfution for 8 hours, 24 hours, 48 hours to simulate myocardial infarction. In this experiment, we used rat cardiac myocytes (H9C2s) for incubation, MTT assay to measure cell viability and Western blot for protein expression.
From the results, the Scutellariae Radix metabolite was capable of protecting hypoxia myocardial cells. It had the effects on cell proliferation, anti-apoptosis, decrease of protein expression of HIF-1α, p-ERK1/2, p-AKT, HO-1, GLUT-4, p-AMPK. From the research, Scutellariae Radix may be used as a Chinese herbal medicine for the prevention of cardiovascular disease.
