Most colorectal carcinoma patients can be treated with surgery and multi-anti-cancer drugs. However, the metastasis of cancer, will result significant decrease in five years survival rate. According to previous findings from Dr. Wang’s lab, the level of adipose differentiation related protein (ADFP) is up-regulated in colorectal carcinoma patients, ADFP is also high expressed in patients with metastasis. Therefore, we speculated that ADFP might contribute to colon cancer metastasis. So far, the relationship between ADFP and cancer is still not clear, so the goal of this study is to clarify the roles of ADFP in colorectal carcinoma metastasis. We used three colorectal carcinoma cell lines to conduct experiments, The low SW480, which has low metastatic ability, SW620 and HT-29, which have high metastatic ability, by manipulating the amount of ADFP expression, we observed the effect of ADFP on cancer cell metastasis. We found that ADFP will promote colorectal cancer cell migration, invasion and sphere formation , but not proliferation, anchorage-independent growth, drug-resistant and epithelial-mesenchymal transition (EMT) capabilities. We concluded that ADFP can promote cell migration, invasion and sphere formation. Yet, it has no significant relationship with EMT and stem cell markers when examined.
