In last years, surgery, radiotherapy (RT) or chemotherapy (CT) can improve the quality of life and reduce the symptom of patients, but the overall five-year survival rate has no significantly increased. In recent advances, cancer stem cells (CSCs) might be the main reason in the therapy failure and cancer recurrence. On the other hand, microRNAs (miRNAs) not only play a key role in development of biological processes but also have the relation to tumor formation. Hence, miRNAs could be a new and effective therapy through regulating gene expression in CSCs. In my studies, I used the flow cytometry to sort CD44+ and CD44- cells, which is CSC marker of head and neck cancer (HNC). And I found that the expression of miR-142-3p in CD44+ cells was down-regulated compared to CD44- cells. Moreover, miR-142-3p over-expression reduced drug resistance by MTT assay and tumorigenic ability in soft agar assay. In addition, miR-142-3p over-expression could suppress tumor growth in vivo. In conclusion, miR-142-3p may play a tumor suppressive role, and hence, miR-142-3p may be considered for therapeutic application for OSCC.
