We have identified several Raman peaks in the spectrum, which can potentially be used to quantify the effect of oxacillin on S. aureus. We used 643 nm diode laser for optical trapping and simultaneous Raman spectra excitation of individual bacterium suspended in liquid. By using different types of measurement, such as time-lapse, continuous, and surface-enhanced Raman scattering we recorded Raman signal from S. aureus with and without oxacillin treatment. Our experimental results show that specific Raman spectral peaks (at 780 cm-1 & 1446 cm-1) are significantly reduced as a result of oxacillin treatment, which can be interpreted as the reduction in the amount of RNA and carbohydrates in the oxacillin-treated bacteria. Under favorable conditions, quantification of the effect of antibiotics on individual bacteria is feasible. Hence, this approach has the potential for drug screening or antibiotics efficacy test with single bacterium
