The treatment of gastric ulcer was considered to be the MMSC(Methionine methylsulfonium chloride) in cabbage. This study was estimated MMSC of extraction of cabbage by various process was analyzed by using HPLC, such as method of squeeze juice(SCE), blended(BCE), extract by (aqueous) RO water(ROCE), 50% ethanol(50%ECE), 95% ethanol(95%ECE) and heat by boiling water(CCE) 30 minutes, finally processed by vacuum concentration and free drying. A comparison contained MMSC between internal leaves and external leaves of cabbage, different storing temperature content of MMSC in cabbage was determined. The result showed that method of SCE obtained the highest content among all of extraction, 50% ECE was the highest content among ethanol extraction and the content of internal leaves was higher than external leaves. In vivo bioactivity study, protection of MMSC on rat gastric mucosal cells, RGM1 cell was observed. In vitro bioactivity study, screening the responsible antioxidant to indentify MMSC with DPPH assays and determined content of polyphenol was studied. The result showed that 95%ECE was the best in antioxidant, the data of polyphenol also consist with the DPPH scavenging rate. By the way, the DPPH scavenging rate was elevated due to D50%ECE and DROCE processed by freeze drying. To scavenging luminol chemiluminescence induced hydrogen peroxide , further to treat RGM1 (rat gastric epithelial cells) cells with those cabbage extracts. The cell vialibility was observed to evaluate the cabbage extract protective effect on gastric mucosal cells. It was also revealed reduced oxidative were follow by concentration dependent. SCE extracted by vacuum concentration was good healing in RGM1 cells, cell viability indicated the effect of protection and antioxidant of MMSC. In this study, we found that MMSC has antioxidant effects, different processed methods enhanced the antioxidant effects of cabbage extract as well, it might be contains other antioxidants such as polyphenols. Analysis of luminol chemiluminescence, inhibition of H2O2 concentration followed by increased of MMSC (concentration dependent), proved that MMSC antioxidant capacity. In the RGM1 cells test, SCE had the best protection. This experiment showed that gastric mucosal protection and antioxidant effect by MMSC or polyphenol, expressing the gastric mucosal damage associated with free radicals. These trials of antioxidant extracts information available to the treatment of future disease research, it was an orientation towards development of gastric mucosal protective agent and antioxidant products.
