Large unilamellar vesicles were prepared first by thin film hydration method, then small unilamellar vesicles were prepared by the sonication method or high-pressure homogenizer method. The freeze-crystal liposomes were prepared by mixing antifreeze(10% sucrose solution)with the liposomes. The purpose of this study is to test liposomes stability, freeze-crystal liposomes stability and their efficacy in skin hydration and to evaluate their safety in human body use. The experimental result revealed that the extraction yield of Auricularia polytricha polysaccharides (A, B, C) were 14.2%;24.8% and 0.8%, respectively. The values of weight-average molecular for samples of A, B and C are 3160±731, 208±36, and 567±83 KDa respectively. Three polysaccharides coated liposomes with the mass ratio of polysaccharides to lipid at 2:1. The encapsulation efficiency in the case of AL was found to be 39.4±14.3%, while 45.3±8.2% for BL and 54.2±12.3% for CL. The average particle size of liposomes produced by the sonication method ranged between 119 nm-182 nm. The average particle size of liposomes produced by the High-Pressure Homogenizer method ranged between 96 nm-119 nm. In regard to liposomes stability, the study period was 28 days, the liposomes stored at low temperature (4℃) exhibited more stability than that were stored at room temperature (25℃). In regard to freeze-crystal liposomes stability, the study period was 8 weeks, the case with antifreeze exhibited more stability than the one without. Freeze-crystal liposomes stored at low temperature (4℃) exhibited more stability than that were stored at room temperature (25℃) and than that were stored at high temperature(45℃). In terms of efficacy, nine formulas that contained Auricularia polytricha polysaccharides (A, B, C) liposome and freeze-crystal liposomes were applied to the skin to test their effects on skin hydration. All formulas were compared with the skin of no tratment but pure solvent. 150 mimutes after the tratment, skin hydration increased in all cases. The rate of change was between 13.2% and 1.1%. Of all, the liposomes style had the best effect on skin hydration.
