Description
Summary:碩士 === 中國文化大學 === 生物科技研究所 === 99 === Taiwan is located in the tropical and subtropical zones. Typhoons or heavy rain in summer season causes flooding, and damages the yield and quality of cauliflower (Brassica oleracea var. Botrytis L.).Cauliflower heat-resistant cultivar ‘H41’, flooding and drought-resistant cultivar ‘H69’, chiling-resistant cultivar ‘H71’ were used in the study. Forty days after transplanting of ‘H41’, 65 days after transplanting of ‘H69’ and ‘H71’, both flooding and non-flooding treatments at 20℃ and 40℃ were conducted. During treatment period, plant growth, SPAD, stomatal conductance, chlorophyll fluorescence Fv/Fm, water potential, and ion leakage percentage of leaves were measured at 0、6、12、24、48、72、96 hr. According to chlorophyll fluorescence, water potential, and SPAD values at 48-hour treatment, ‘H41’ plants showed better flooding and heat tolerance than others. The leaves of 6- and 24-hr treated plants were then used for proteomic analysis. For ‘H41’, there were 6, 5, 10, 1, 6, and 13 peaks with different protein expressions in leaves of non-flooding treatment at 24 hr and 20℃, flooding treatment at 6 and 24 hr and 20℃, non-flooding treatment at 6 and 24 hr and 40℃, and flooding 24 hr at 40℃, resrectively. For ‘H69’, there were 10, 3, 15, and 15 peaks with different protein expressions in leaves of non-flooding treatment at 20℃, flooding treatment at 20℃, non-flooding treatment at 40℃and flooding treatment at 40℃ for 24 hours, resrectively. For ‘H71’, there were 5, 1, and 5 peaks with different protein expressions in leaves of non-flooding treatment at 20℃, flooding treatment at 20℃, and flooding treatment at 40℃ and 24 hrs, resrectively. These proteins are related with cell membrane stability, amino acid biosynthesis, photosynthesis, amino acid translation, respiratory metabolism, and fatty acid synthesis.
