It has been reported that Subamolide B has been shown to be cytotoxic to human colon cancer. However, the mechanisms of Subamolide B inducing cancer cell death and anti-cancer ability remain unclear. Here we used SCC-12 which is a skin squamous cell carcinoma as a cell model to study the mechanisms of Subamolide B. In this study, I found that the cell viability was significant decreased after Subamolide B treatment, besides, the cell colony formation ability was inhibited. Furthermore, Subamolide B induced endoplasmic reticulum-stress ( ER-stress ) generation in cancer cell and activated two apoptotic pathways including intrinsic and extrinsic pathways through up-regulation of Fas ligand and down-regulation of Bcl-2. Additionally, Subamolide B do increase the phosphorylation of JNK, ERK, and p38 all in dose- and time-dependent manners. Meanwhile, caspases cleavage-form decreased after MAPKs inhibitor treatment. Importantly, Subamolide B regulates apoptosis and related proteins expression through ERK and JNK pathways. Moreover, SCC-12 cell has G2/M phase arrest after Subamolide B treatment. Overall, in this study, we demonstrated that Subamolide B could induce cell apoptosis, decrease colony formation ability, and induce G2/M phase arrest in skin squamous cell.
