In the previously study, the partcial amino acid sequence of weissellcin 110 has been illustrated, however, the full-length sequence of this bacteriocin is still lacking. In current thesis, the attempt to discovery the DNA sequencing of both flanking regions neighboring the known sequence region, that included the 5’ and 3’ sequence unknown regions of wessellcin 110, and the genes neighbored. Two strategies that (1) traditional inversed PCR with restriction enzyme and T4 DNA ligase pretreated circular genomic fragmented template (2) newly designed PCR with random and ATCG sequence -based linker primer was introduced to deal with the problem. The current data demonstrated that both two strategies are work to amplify a constant PCR product. The following cloning and sequencing experiments are proceeding.
