This study investigates the physicochemical properties of heparin and nerve growth factor (NGF)-grafted polycaprolactone (PCL)/poly(3-hydrobutyrate) (PHB) scaffolds and the NGF-guided differentiation of iPSCs in the scaffolds. The PCL/PHB scaffolds were fabricated by solvent merging/particulate leaching method. The PCL/PHB scaffolds with the mass ratio of 4:1 have a relatively high porosity and cell adhesion. In addition, an increase in the weight percentage of heparin enhanced the cell viability. The flow cytometry analysis of heparin and NGF-grafted PCL/PHB scaffolds demonstrated that when NGF was introduced, an increased weight percentage of PCL yielded more identified neurons. In addition, the immunocytochemical staining revealed that PCL/PHB scaffolds grafted with heparin and NGF could induce the differentiation of iPSCs into neurons via the expressed β-III tubulin. A proper control over the physical and biomedical property of the scaffolds and the NGF-guided differentiation of iPSCs can be promising for nerve regeneration.
