Studies on the molecular events indicated that PICK1 suppressed etoposide-induced dimer formation of the pro-apoptotic protein Bax in human leukemia REH cells. In addition to etoposide, PICK1 also suppressed doxorubicin-induced cell death but had little effect on cisplatin-induced cell death. In this study I showed that PICK1 also suppressed doxorubicin-induced but not cisplatin-induced Bax dimerization, suggesting that suppression of Bax dimerization is a common molecular event through which PICK1 suppressed drug-induced killing. I also used GST-PICK1 wild type and mutant recombinant proteins to pull down potential interacting proteins. Further identification of interacting proteins will help examining the function of PICK1.
