Ectopic expression of PICK1 in leukemia REH cells affects cellular response to chemotherapeutic agents

(PKC?? to mitochondria and thus stabilized mitochondrial membrane potential upon etoposide treatment. Studies on the molecular events indicated that PICK1 suppressed etoposide-induced dimer formation of the pro-apoptotic protein Bax in human leukemia REH cells. In addition to etoposide, PICK1 also suppressed doxorubicin-induced cell death but had little effect on cisplatin-induced cell death. In this study I showed that PICK1 also suppressed doxorubicin-induced but not cisplatin-induced Bax dimerization, suggesting that suppression of Bax dimerization is a common molecular event through which PICK1 suppressed drug-induced killing. I also used GST-PICK1 wild type and mutant recombinant proteins to pull down potential interacting proteins. Further identification of interacting proteins will help examining the function of PICK1.
