Description
Summary:碩士 === 東海大學 === 化學系 === 98 === Dipeptide compounds play an important role for complex physiology functions in the human body. They suppress mainly the carbonylation, carbohydrate, and oxidation reaction to avoid the human ageing. In this study, we establish a set of analysis method for the detection of six kinds of dipeptide compounds by using MALDI-TOF-MS. The molecular weight of these dipeptides are below 250 Da. Using traditional organic acids as matrix, the mass spectrum will present low mass interferent background and make the analyte signal unable to be resolved. To solve the matrix interferent problem, we use nano colloidal gold to replace organic acid as matrix. It can effectively reduce the matrix background, and successfully applys to analyze the six dipeptide compounds. Our experimental results show the detection limit is between 2.23 ppm to 5.78 ppm, and the linear correlation coefficient (R2) is more than 0.99. The advantages of this method are easy operation, large number of sample analysis, fast and simple procedure, high sensitivity and high resolution. This method can be applied to determine the dipeptide compounds in urine sample.
