Laetiporus sulphureus is an edible mushroom known to cause heart-rot disease in deciduous trees and conifers. It is found in the broad-leaved tree trunk of low elevation region in Taiwan. The optimum cultural medium and some bioactivities such as antitumor for water extract from L. sulphureus had been studied in our Lab. Therefore, the purpose of this study is about the biological effects of the alcohol extract of mycelium from L. sulphureus (PM-E, SC-E) enhance Doxorubicin(DOX)-induced cytotoxicity in breast carcinoma cell lines. The inhibitory effect on the breast cancer cell’s (MCF-7 and MDA-MB-231) growth, antimetastasis experiment and the antioxidation experiments were conducted. The results showed that the alcohol extract of mycelium from L. sulphureus could significantly inhibit the growth of breast carcinoma cells due to MTT-assay. Moreover, the mechanism of the growth-inhibition was due to apoptosis pathway evaluated by flow cytometer. On one hand, the alcohol extract of mycelium from L. sulphureus significantly enhanced Dxorubicin( DOX ) -induced cytotoxicity and also affected the cell cycle in breast carcinoma cell lines. The results have shown that the alcohol extract of mycelium from L. sulphureus could modulate the protein expression of Bax、Bcl2、Caspase-3、Caspase-9 in RT-PCR.The anti-metastasis was conducted by getalin zymograph and the results showed that the secretion of protein were obviously suppressed while HT1080 cells were treated with the alcohol extract of mycelium from L. sulphureus. In the regulation of the immunity, only PM-E treat MCF-7 and MDA-MB-231 had effect to promote the immunity in the statistics. The results showed that the alcohol extract and the water of mycelium from L. sulphureus could modulate the cytokines expression of IL-1、IL-6、TNF-α in RT-PCR.These results suggest that the alcohol extract of mycelium from L. sulphureus has some bioactivities such as antitumor , immunomodulatory and antimetastasis effects.
