Summary: | 碩士 === 實踐大學 === 食品營養與保健生技學系碩士班 === 98 === Auricularia auricula-judae (AAJ) is one kind of vegetable that rich in dietary fiber, vitamins and various enzymes, also content high level of iron and calcium. Recently studies have shown AAJ contain anticoagulant substances to lower blood viscosity, prevent thrombosis and reduce the risk of atherosclerosis. Our experiment was crushed AAJ into powder then extracted with distilled water and ethanol individually from 2 to 12 hours evaluate the extraction yield, antioxidant activity and angiotensin I-converting enzyme (ACE) inhibition in vitro. After that we began animal experiments to test changes in blood pressure during every two weeks. In the end of the experiment we measured the different of plasma and organs of the ACE inhibition activity. The results showed that distilled water extraction yield dramatically higher (4~10%). In antioxidant activity tests, AAJ distilled water extract (AWE) and AAJ ethanol extract (AEE) the IC50 values in DPPH scavenging respectively AWE 836~968μg/mL, AEE 778~890μg/mL and in reducing power AWE 919~1357μg/mL, AEE 773~850μg/mL and ferrous ion chelating effect AWE 918~1933μg/mL but no significant effect of AEE. AWE the ACE inhibition in vitro and the IC50 values was 11~44μg/mL. Upon previous results we picked 2 hour cold distilled water extract (2CWE) further to feed SHR rats to evaluate the blood pressure for 8 weeks. Divided SHR rats into high, medium, low dose group (1000, 500 and 200 mg/ Kg BW/day in drinking water), AAJ powder replace 5% of the feed formula group, SHR rats-control (basal diet) and SHR rats-positive control (basal diet contain 20 mg/Kg BW/day Captopril ) group. Result showed at 2 ~ 8 weeks and 4~ 10hrs at 7weeks, systolic blood pressure of low, medium and high dose group showed significantly lower than SHR-control group ( p < 0.05 ). ACE activity of plasma, liver and kindy were significantly lower then SHR-control group.
