The E-isomer is known as geranial or citral A. The Z-isomer is known as neral or citral B. There are several antioxidant-response element (ARE)-regulated gene products such as glutathione S-transferase, NAD(P)H:quinine oxidoreductase 1, glutamate cysteine ligase, and hemeoxygenase-1 are known to mediate detoxification and to exert antioxidant functions thereby protecting cells from genotoxic damage. The transcription of ARE-driven genes is regulated, at least in part, by nuclear transcription factor erythroid 2p45 (NF-E2)-related factor 2(Nrf2), which is sequestered in cytoplasm by Kelch-like ECH-associated protein 1 (Keap1). This experiment will investigate the effects of citral on the glutathione (GSH) production, phase II detoxifying enzymes protein expression, Nrf2 protein levels and translocation and it regulatory pathways. In the results, concentration of citral under 100 μM showed no toxic effect on Hep G2/C3A cell lines. We found that the pathway of citral activation of Nrf2 through IV phosphorylation by Akt, JNK and ERK. We also found that increase in Nrf2 accumulation in the nuclear fraction, induce phase II detoxify enzyme, including GCLC, GST and HO-1 and induction of GSH synthesis.
