Summary: | 碩士 === 國防醫學院 === 藥學研究所 === 98 === Tangshen, the root of Codonopsis pilosula. ( Franch.) Nannf., is a common used drug in traditional Chinese medicine. In clinical, tangshen used to reinforce qi and usually replace ginseng.
The 95% alcohol extract of the root of C. pilosula. was dissolved in water and according to partitioned with n-hexane, dichloromethane, ethyl acetate and n-butanol to give five layers, COPI-H, COPI-D, COPI-E, COPI-B and COPI-W. The COPI-H, COPI-E and COPI-B layers were chromatographied through Diaion HP-20, silica gel and reversed phase chromatography, to get nine compounds, adenosine (1), hexyl β-sophoroside (2), butyl-β-D- fructopyranoside (3), feridelin (4), taraxerol (5), Olean-12-en-3β,15α-diol (6), lobetyolin (7), cordifolioidyne B (8) and codonolin C (9).
Among them, Olean-12-en-3β,15α-diol (8) is first time isolated from C. pilosula and codonolin C (9) is a new compound.
The Caco-2 cell line were used as bioactivity experimental models to measure the glucose uptake. The glucose uptake could be increased by COPI-E, COPI-B and COPI-W. But isolated compound have no remarkable effect. The activity compounds were remains to be clarified.
