spelling ndltd-TW-098NCYU57300072015-10-13T18:35:12Z http://ndltd.ncl.edu.tw/handle/27882356078820489784 The Study on Microfluidic System for Cell Lysis and DNA Extending 細胞裂解與DNA拉伸微流體系統之研究 Chih-Yi Liu 劉志益 碩士 國立嘉義大學 生物機電工程學系研究所 98 This study uses the mode of a closed channel by integrating the microfluidic devices and the techniques of chemical cell lysis with the micro-peristaltic pump into the manipulating worktops,which facilitates the instant observations of cell lysis and DNA extending。The design of the microfluidic devices is composed of three parts:the first part is cell separation;the second part is cell fixation;and the third part is DNA extending。The result of this study is that the numbers of the lysed cells can be decreased by the separation of the two Y-type bifurcated channels and fewer cells flow into the cell fixation regions by the micropump, when the cells are initially injected into the entrance of the microfluidic model into the closed channels。 Using the designed microfluidic channel, the cells can be fixed through infusing cell fixing reagent Cell-Tak solution in front of the rectangular structure。 When the cells are fixed on glass substrate and lysed withe the SDS(1%) cell lysing solution,the DNA of lysed cells are released into the solution the fluorescent dyed DNA extending can be observed.
