Needle-free injection system is two sections of step. The tissue is first penetrated into the mucosal layer and lifted up due to the second-step injection of the working fluids into the tissue. It is then recovered due to the diffusion of the working medium to the neighboring tissue. The working fluids include normal saline glycerin and alginate solutions. Results show that the alginate solution with higher rheology property has the advantage to supply higher flow rate under the same condition. Results also show that when using normal saline as the injection fluid at the first stage. The best injection parameter is injection is injection power of 0.5 W and above. Injection of power more than 0.5 W is ensured to penetrate into the mucosal layer. The second stage jet injection power of 0.1 W and above. Injection of power more than 0.1 W is ensured to carry out tissue elevation. The descending time of alginate and glycerin solution with viscosity of 5cp reduced by 50 % (define as the half-life time of the tissue elevation) is exceeding 60 and 30 minutes. Normal saline has the shortest half-life time of about 18 minutes. The porcine colon used alginate solution to tissue elevation can separate effectively the carbon dioxide snow.
