The effects of solvent concentration and solid to liquid ratio on flavonoids yield were individually determined. The optimum conditions of hot pressure water extraction for flavonoids yield were determined pressures at 1400 psi, 5 min extraction time and temperatures at 55 ℃. The yield of flavonoids obtained by hot pressure water extraction is 16.04 mg/g, and it was higher than there from ultrasonic method (7.91 mg/g), Thus, Hot pressure water extraction has higher efficiency of extraction and required less extraction time compared to other extraction methods. The quantitative analysis of five flavonoids were determined by LC/MS2. By optimizing the chromatographic separation and mass spectrometric parameters, the precursor ions of the derivatives with the best responses were found. The optimal ionization characteristics were obtained when using electrospray ionization (ESI) in negative-ion mode. The sekected reaction monitoring (SRM) mode provided additional improvement in selectivity. The analysis time including column equilibrium was 30 min per sample. The limits of detection (LODs) for K1, K2, N1, N2, and N3 were 43.4, 26.9, 37.8, 44.3, 19.5 ng/mL, respectively. The linearity range was 100-2000 ng/mL. 鍾婷婷 學位論文 ; thesis 80 zh-TW
