In recent years, many papers pointed out that the beneficial to human, including hypolipidemic, hepatic protection functions, functional ingredients which contains cordycepin, adenosine, polysaccharide, mannitol and superoxide dismutase. However, natural C. sinensis is a limited production, coupled with gradually increasing market demand, study of artificial cultivation of C. militaris driven, and pharmacological effects is similar of C. sinensis, and the prices are much cheap. The purpose of this thesis was to study the improvement of medium composition in order to promote the growth of C. militaris, reproduction and the synthesis of bioactive substances, mainly in solid medium, using rice as their main medium. The solid cultivation medium is the rice 49.4%, distilled water 49.4%, amino acids 0.2% (15 amino acid individual added) and 1% acetic acid solution of chitosan (chitosan-containing 0.3%), cultivated conditions as follows: temperature 20 ~ 22°C, relative humidity 70 ~ 80% for 60 days cultivation, harvesting of fruiting body in C. militaris, record the number of trees, length and the weight, then vacuum freeze-drying. For quality analyzed, color is analyzed by colorimeter, cordycepin content is analyzed by high-pressure liquid chromatography (HPLC), polysaccharide molecular weight is analyzed by gel permeation chromatography (GPC) from fruiting bodies of C. militaris, polysaccharide content is analyzed by phenol-sulfuric acid from fruiting bodies of C. militaris, mannitol content is analyzed by oxidation-reduction titration. The results showed that adding different amino acids and 0.3% chitosan in acetate solution treatment, cordycepin content more than added chitosan, but the length and yield of fruit body are opposite. The addition of 9 group amino acids(including alanine、valine、isoleucine、tyrosine、tryptophan、histidine、serine、threonine and arginine) show morecontents of cordycepin in fruit bodies, and the synthesis, fruiting bodies than the controlled group. The sample of valine added show the highest of cordycepin content is 11.56 mg/g (dry matter). The sample of threonine added show the highest of polysaccharide content is 23.70 mg/g (dry matter). Arginine group of basic amino acids at single added, yield, length and cordycepin content is more than adding 0.3% chitosn in acetate solution of. The sulfur-containing amino acid cysteine at single added. There are no growth of fruit bodies during culture, beside adding 0.3% chitosan in acetate solution.
