Summary:碩士 === 輔仁大學 === 食品科學系 === 98 === The objectives of this study were to develop a high performance liquid chromatography-mass spectrometry method for determination of carotenoids and chlorophylls in R. nasutus, a tradition herb possessing anticancer and antioxidant activities, as well as study their variation upon drying treatment. Identification of both carotenoids and chlorophylls was carried out on the basis of retention behavior, absorption spectra and mass spectra using atmospheric pressure chemical ionization (APCI) in positive ion mode for detection. Results showed that a total of 24 carotenoids were separated within 54 min by using a Waters YMC C30 column and a gradient mobile phase of methanol-acetonitrile-water (82:14:4, v/v/v) and methylene chloride with flow rate at 1 mL/min and detection wavelength at 450 nm. Likewise, a total of 12 chlorophylls were separated within 21 min by using a Agilent Eclipse XDB-C18 column and a gradient and mobile phase of MeOH- N,N-dimethylformamide (97:3,v/v), acetonitrile and acetone with flow rate at 1 mL/min as well as detection wavelength at 660 nm. The highest yield of total carotenoids and chlorophylls were achieved by freeze-drying, however, the isomerization and degradation of both pigments were more pronounced with hot-air drying than with freeze drying.
