Some researches indicate that consumption of soymilk decreases cardiovascular disease risk, such as hypertension and atherosclerosis. In this research, soymilk fermented with lactic acid bacteria isolated from Taiwanese pickles and sourkaurts, Lactobacillus plantarum TWK10 and Pediococcus pentosaceus MK6, were used to evaluate the improvement of hypertension and atherosclerotic effects in cell culture model system. Moreover, the supernatants, water extracts and ethanol extracts were determined the contents of peptides and isoflavones. Then, the angiotensin converting enzyme (ACE) inhibitions were also investigated. As the result, the soymilk fermented by L. plantarum TWK10 and S. thermophillus BCRC14085 for 48 hours had the higher production of peptides and transformation of glucoside isoflavones to aglycone isoflavones than P. pentosaceus MK6. In inhibition of ACE activity, the IC50 value of the water extracts from soymilk fermented with L. plantarum TWK10 was significantly (p<0.05) lower than S. thermophillus BCRC14085 and P. pentosaceus MK6. The ethanol extracts of fermented soymilk stimulated higher production of nitric oxide than water extracts in human umbilical vein endothelial cells (HUVEC). Moreover, the nitric oxide production which were stimulated by the extracts of soymilk fermented with L. plantrum TWK10 and S. thermalphillus BCRC14085 were significantly (p<0.05) higher than P. pentosaceus MK6. Furthermore, ethanol extracts of soymilk fermented with L. plantrum TWK10 and S. thermalphillus BCRC14085 showed higher scavenging effect of superoxide anion and stimulating effect of prostaglandin E2 (PGE2) production than water extracts. Western blotting analysis of endothelial nitric oxide synthase (eNOS) in HUVEC treated with ethanol extracts were higher than water extracts. In addition, fermented soymilk increased inhibitory efficiency of proliferation, migration and enhanced the mRNA expression of E-prostanoid 4 (EP4) receptor in rat thoracic aortic smooth muscle cells (A10 cells). In summary, fermented soymilk was found to improve endothelial dysfunction, ameliorate hypertension and atherosclerosis.
