The operating parameters of the response surface experiment are green tea powder, selenium yeast, and glucose of the corresponding viscosity, to identify the optimal package of meal processing conditions and nutritional composition. The clinical trial subjects will be screen and assigned according to the following conditions: the body mass index higher than 27, randomly assigned to the three groups: control group, green tea group and the chitosan group;a total of 45 subjects will participate the 12-week weight control program to compare the difference in their anthropometric measurements values, biochemical values and the values in physiological responses. The testing results indicate that the optimum operating parameters condition is under the green tea powder: 5.4 ± 0.7 g, selenium yeast: 1.3 ± 0.2 g, glucose powder 7.5 ± 0.2 g. The closest to the design value of the formulation viscosity 78 CPS (p <0.005), 136 ± 5 kcal per pack, and net weight 17.5 ± 0.5 g can be obtained. Meanwhile, the clinical trial results, in body weight, waist circumference, low-density / high-density lipoprotein ratio change, have a significant change in green tea group and in the chitosan group, the data are 4.7 ± 1.4 and 2.3 ± 0.5 (kg), 7.5 ± 0.4 and 3.2 ± 1.5 (cm), 0.69 ± 0.12 and 0.26 ± 0.32 (mg / dL). The data shown that green tea group has much significant than chitosan group, while in the control group did not have the significant difference. However, in physiological responses, the insomnia condition that happened in green tea group shown the significant different compare to chitosan group, but had no significant difference in chitosan group and in the control group. Because of the physiological responses in green tea group has the significant difference compare to the other groups, thus we added another physical fitness group in clinical trials. The experimental conditions are same as before and we compare the data with the green tea group. The results indicate that the body weight and waist circumference in physical fitness have a clearly difference in the green tea group, were 7.9 ± 3.4 (Kg) and 8.9 ± 1.4 (cm). The physiological response on the physical fitness group that caused by hunger condition is significant compare to the green tea group. Through the response surface method, the appropriateness of the estimate values in each response characteristics of the regression models and the actual measured values in the product can be confirmed. The clinical trials also validate that self-made weight-reducing meal replacement has the effect in lowering the body weight and the blood lipid.
